This DNA/RNA Clean-up and Concentration Micro Kit offers a fast and efficient method for purifying, cleaning up, and concentrating DNA or RNA from various isolation techniques or upstream processes. It is capable of purifying all sizes of DNA, ranging from small PCR products and native or linearized plasmids to genomic DNA. The kit serves as a convenient alternative to organic extraction and ethanol precipitation for cleaning up different enzymatic reactions. It effectively removes PCR byproducts such as primers, primer dimers, enzymes, and unincorporated nucleotides. The purified DNA is fully compatible with downstream applications including restriction enzyme digestion, ligation, labeling, PCR, and sequencing.
In addition, the kit efficiently purifies RNA obtained from phenol/guanidine based extraction protocols or from enzymatic reactions such as DNase treatment, labeling, and in vitro transcription. It isolates all RNA sizes from large mRNA and ribosomal RNA to microRNA (miRNA) and small interfering RNA (siRNA) preferentially removing proteins, nucleotides, and other contaminants without using phenol or chloroform. The resulting RNA is highly intact and suitable for a variety of downstream applications including end-point or quantitative reverse transcription PCR, Northern blotting, RNase protection assays, primer extension, and expression array analyses.
Furthermore, the kit includes a protocol for rapid endotoxin removal from previously purified DNA, reducing endotoxin levels to 0.1 EU/μg DNA or less. Endotoxins, also known as lipopolysaccharides, are components of the outer membrane of Gram-negative bacteria such as E. coli. These endotoxins are released during bacterial cell lysis in plasmid purification and can significantly impair transfection efficiency in endotoxin-sensitive cell lines. Therefore, removing endotoxins from plasmid preparations is essential before using the DNA in sensitive downstream applications.
